dc.description.abstractHemicellulose is a component of Non-Starch Polysaccarides(NSP) contained in agricultural byproduct and can be separated from the other components of NSP by isolation or hydrolysis. Various studies about hemicelluloses isolation procedures from various agricultural byproduct material have been conducted including corncobs, and also to use of hemicelluloses in the pharmaceutical field as drug release systems, anti-oxidant and anti-microbial. Effect of cadmium and lead exposure in humans, such as anemia, hypertension, autism, cardiovascular, renal dysfunction, carcinogenic, encepalopati and peripheral neuropathy has been conducted by many researchers. In connection with the above it is necessary to study the natural materials that can bind to the metal. Various studies have been conducted on the use of chitosan, sodium alginate and pectin to bind iron, alminium, copper and lead. hemicellulose has hydroxyl, carbonyl, and carboxylic possibilities can be used to bind metals cadmium and lead in vitro and in vivo. Objective of this research his to modify the isolation hemicelluloses from corncobs, also corncobs hemicelluloses (CCH) to bind cadmium and lead ions in alkaline and acidic pH also the corncobs hemicelluloses (CCH) to bind cadmium and lead in vivo by parameter cadmium and lead in the blood and liver function and kidney function. This research includes isolation hemicellulose from com cobs using 3 procedures that have been done before and the researcher is a modified procedure conducted by researchers themselves. Characterization CCH done by infrared spectrophotometry (FTIR), high performance liquid chromatography (HPLC) with column Shim-pack VPODS (4.6 x 250 mm) using a UV-Vis detector at a wavelength of280 nm with an isocratic elution system, solvent aquabides, and a flow rate of 0.8 mL / min. binding ability CCH done 3 ways: first, by titrimetric with cadmium 3 mg / ml and 10 mg of lead / ml as a titrant and indicator are 0.05 N NaOH, and the second CCH binding ability in vitro at pH 2, as a comparison to use pectin. Third Test in vivo conducted 3 variations of treatment that CCH IO mg, 100 mcg and I 00 mcg of lead cadmium for IO weeks. Assay of cadmium conducted using atomic absorption spectrophotometry with flame at a wavelength of 228.8 nm and lead using atomic absorption spectrophotometry with a graphite furnace at a wavelength of283.3 nm. The results showed that the modification of isolation, provide the greatest yield CCH is 12.04%, with characteristic FTIR region 1820-1600 cm-, vibration widened near 3800-3000 cm-1., Meaning the carbonyl group, hydroxyl and carboxylic groups on the CCH structure. The results showed that the yield of HPLC chromatograms CCH has a retention time of 1.8 minutes. Test results of the titrimetric holding capacity, that CCH was binding 100 mg of cadmium (46.17 ± 0.9256) mg or 46.17%, and lead (65.99 ± 1.4280) mg or 65.99%. Holding capacity test results at pH 2 showed that 300 mg of CCH yield was binding 30 mg cadmium of (26.68 ± 0.1490) mg or 88.93%, and 40 mg lead of (39.52 ± 0.1350) mg or 98.80%. The results of in vivo tests showed that cadmium levels decreased by 95.05%, and 94.81 % for lead, and had no effect on the parameters of liver function and kidney function. Based on the exposure it can be concluded that the modification produces CCH isolation yield of 12.04% and can reduce to cadmium and lead levels in the blood without affecting the function of the kidneys and liver.en_US
