Background. Tuberculosis (TB) is an infectious disease caused by bacteria (Mycobacterium tuberculosis) which often affects the lungs. Gene polymorphism is one of many risk factor for susceptibility of lung tuberculosis that have been proven in genetic studies, one of which is the TLR4 Asp299Gly gene polymorphism. Aim. Determine the association between TLR4 Asp299Gly gene polymorphism with lung tuberculosis susceptibility. Methods. Case-control study performed in 60 newly diagnosed lung tuberculosis cases patient and 60 healthy control group in Medan. TLR4 Asp299Gly gene polymorphism analysis with PCR-RFLP procedure and electrophoresis in agarose 4%. Results. TLR4 Asp299Gly gene polymorphism unsignificantly increased allele and genotype frequency in control group compared to case group [G Allele: 2,5% vs 0,8%; p = 0,31; OR (95% CI) =3,05 (0,31 – 29,75)] [AG Genotype: 5% vs 1,7%; p = 0,31; OR (95% CI) = 3,11 (0,31 – 30,73)]. Conclusion. There were no statistically significant differences in both distribution of genotypes and alleles, it can be concluded that the TLR4 Asp299Gly gene polymorphism is not a risk factor for pulmonary tuberculosis in this study.en_US
