Introduction. Staphylococcus aureus is a bacterium that causes frequent occurrences of blood vessel infections and over time this bacterium can mutate into Methicillin-Resistant Staphylococcus aureus. One way to prevent this is to use natural-based antibacterial agents, one of which is the Moringa oleifera plant. It is known that secondary metabolites in Moringa oleifera leaves have antibacterial potential. In addition, the harvest time is also estimated to have an influence on the amount of secondary metabolites contained in the plant. Objective. Knowing the antibacterial effectiveness of Moringa oleifera extract against Staphylococcus aureus ATCC 25923. Method. quasi-experimental research design with posttest only control group design. The extract is made by maceration. There are 3 concentration groups (60,70,80%) and 2 harvest time (morning and evening). Testing using diffusion method with 4 repetitions with measurement parameters of the length of the diameter of the inhibition zone. The data were analyzed by Friedman Test and followed by Post-Hoc test. Results. The diameter of the inhibition zone of P80 against Staphylococcus aureus (10,375- 0,144 mm) was longer than the other morning group. The diameter of the inhibition zone of S80 against Staphylococcus aureus (10,687-0,473 mm) was longer than that of the afternoon group. There was a significant difference between the treatment groups (p < 0,05). Conclusion. 96% ethanol extract of Moringa oleifera leaves has the ability to inhibit the growth of Staphylococcus aureus ATCC 25923.
