dc.description.abstractIsolation of protease enzymes from papaya leaf extract (Carica papaya L) and enzyme purification has been conducted by precipitation method with ammonium sulfate at a concentration of 40-60% and dialysis with cellophane membrane was then characterized before and after immobilization using polyacrylamide gel to determine the optimum conditions. The protease enzyme activity was determined using a UV-Visible Spectrophotometer. The optimum condition was obtained from crude extract of protease, free protease enzyme, and immobilized protease enzyme from papaya (Carica papaya L) leaf extract were at pH 7, temperature reaction 40oC and reaction time of 40 minutes with the activity 11,2288; 12,7992 and 11,9018 U/ml. The protease enzyme that has been immobilized with polyacrylamide gel can be used repeatedly 4 (four) times and still shows an efficiency of 74,75% on the fourth use.en_US
