Isolasi dan Identifikasi Golongan Senyawa Fenolik dari Kulit Batang Tumbuhan Kalpataru (Hura Crepitans Linn.)

Isolation and identification of groups of phenolic compound contained in the bark of the Kalpataru Plant (Hura crepitans Linn) have been carried out by maceration extraction using methanol as a solvent. The concentrated methanol extract was re-extracted using ethyl acetate repeatedly until it was negative for FeCl3 5%. The ethyl acetate extract was concentrated and redissolved with methanol and then partitioned with n-hexane until the n-hexane layer was clear. The methanol extract was separated by column chromatography with a stationary phase of silica gel and a mobile phase of chloroform: ethyl acetate (90:10; 80:20; 70:30; 60:40 50:50; 40:60) v/v, respectively. The compound obtained from fraction 131-190 was purified by preparative thin layer chromatography to produce 12 mg yellow amorphous solid with an Rf value of 0,44 with chloroform: methanol (90:10) v/v as the eluent. Analysis of the compounds obtained was carried out using UV-Vis spectrophotometry, FTIR, and 1H-NMR. The UV-Vis spectrum shows a wavelength (λ max) of 217 nm in band II and 275 nm in band I. The FT-IR spectrum shows the presence of –OH stretching, CH stretching, C=O stretching, C=C stretching, bending C-H, C-O stretching, respectively, at wave numbers 3356,14 cm-1 , 2924.09 cm-1, 1693,50 cm-1, 1535,34 cm-1, 1442.75 cm-1, 1002,98 cm-1. The 1H-NMR spectrum shows the presence of protons H-2 and H-6 in the aromatic ring of phenolic compounds, protons H-2' from CH3 and protons H-4 from CH3 attached to the benzene ring and the H-1' of OCH2 and protons H from CH2 which uses on the aromatic ring. Based on the data obtained, it is suspected that the isolated compound obtained is a phenolic compound derived from phenylacetic acid.
