Recycling cooking oil for deep frying makes the transformation of certain unsaturated fatty acid from cis to trans configuration. There is a direct effect of trans fatty acid on Nonalcoholic Fatty Liver Disease (NAFLD). NAFLD is the most common form of liver disease in the general population of both Western and Eastern countries. The incidence of NAFLD has increased rapidly due to the ongoing epidemics of obesity and type 2 diabetes. However, there are no specific drugs to prevent this disease. This study aimed was to perceive the hepatoprotective effect of Pleurotus ostreatus. This research was an experimental study using randomized control group post test only design. Subjects of this research were 32 male Wistar rats and divided into four groups (n=6 rats/treatment). Group PO just received aquadest. Group Pl as positif control received deep frying oil (0,42ml/200 g body weight/day). Group P2 and P3 received deep frying oil (0,42ml/200 g body weigh.tlday) and ethanolic extract of Pleurotus ostreatus (respectively 200mglbody weight/day and 250mglbody weight/day). Measurement of Alanin Transaminase (ALT) level and calculation of lipid droplets were done at 57 day after treatment. Data from this research was analyzed using Kruskal-W allis. There were significant difference among four groups (p = 0, 000) for ALT serum level and lipid droplets quantity. The result of Post Hoc Test for ALT serum level and lipid droplets quantity showed that there were significant difference between group PO with group Pl, and between group P2 with group Pl. Ethanolic extract of Pleurotus ostreatus reduced the development of NAFLD by reducing liver steatosis and ALT serum level. This research showed that Pleurotus ostreatus could prevent NAFLD in wistar rats with effective dose 200mglbody weight/dayen_US
