Chemo-mechanical preparation procedures on root canal treatment is also called forming (shaping) and cleaning (cleaning) which uses file and irrigation materials in shaping and cleaning the root canal of a smear layer Protaper Universal design and iRace file rotary different can generate variations in the cleaning efficiency of smear layer in the root canal. Irrigation material that is generally used is Sodium Hypochlorite (NaOCI) are toxic and often cause allergies while EDTA is one of the ingredients of irrigation effectively remove smear layer is formed after the preparation but EDTA does not have antimicrobial effects. Chitosan as natural polysaccharide after cellulose obtained by deacetylation of chitin have biocompatible properties, bioadhesi and no toxins in human cells. This study aimed to see if there was an effect of use of Protaper Universal and iRace file rotary with i irrigation material NaOCI 2.5%, EDTA 17% and 0.2% high molecular chitosan to cleanliness intracanal smear layer on the apical third channel roots and whether there are differences in the effect of the use of Protaper Universal and iRace rotary file with irrigation material NaOCI 2.5%, EDTA 17% and the high molecular chitosan 0.25% against hygiene intracanal smocar layer at the apical third of the root canal. Root canal preparation was performed on 32 samples of mandibular premolar teeth were divided into 8 groups. The first, second, third and fourth group using Protaper Universal file rotary with irrigation materials NaOCI 2.5% and EDTA 17% NaOCI 2,5%, NaOCI 2.5% and High Molecular Chitosan 0.2% and High Molecular Chitosan 0.2% The fifth, sixth, seventh, eighth group using iRace file rotary with irrigation materials NaOCI 2.5% and EDTA 17%, NaOCI 2,5% NaOCI 2.5% and High Molecular Chitosan 0,2% and High Molecular Chitosan 0,2%. Kruskal-Wallis test showed no significant effect of universal Protaper with irrigation material NaOCI 2.5%, EDTA 17% and 0.2% high molecular chitosan to cleanliness intracanal smear layer at the apical third of the root canal (P>0.05), but a significant difference using a iRace file rotary (P <0.05). Mann-Whitney test showed no significant difference in each group ProTaper universal and irrigation materials Irace with NaOCI 2.5%, EDTA 17% and 0.2% high molecular chitosan to cleanliness intracanal smear layer on apical third of the root canal (P>0.05), but when viewed from the median value of Protaper Universal and Race rotary file with irrigation materials of high molecular chitosan 0.2% more effective in cleaning the smear layer on the apical third of the root canal. In this study it can be concluded that the Protaper Universal and iRace file rotary with irrigation materials of high molecular chitosan 0.2% more effective in cleaning the smear layer on the apical third of the root canal irrigation materials compared with NaOCI 2.5% and EDTA 17%
