Plaque is a soft layer consisting of a collection of microorganisms. Streptococcus sanguinis is a pioneer bacterium that forms a colony and plays a major role in the formation of oral biofilms. Prevention of plaque can be done by mechanical or chemical action using mouthwash. Red dragon fruit skin (Hylocereus polyrhizus) contains antibacterial compounds such as alkaloids, flavonoids, saponins, and triterpenes/steroids. Chitosan can function as an antibacterial because it contains polysaccharide amino groups so it has the potential to be combined into mouthwash. This study aims to determine the effectiveness of mouthwash containing 0.5% chitosan solution and 25% red dragon fruit peel extract in inhibiting the growth of S. sanguinis bacteria. This type of research is a laboratory experimental research with a post-test-only control group design research design. The sample for this study was a pure culture of S. sanguinis ATCC®10556TM which was then tested for antibacterial effectiveness using the diffusion method (Kirby-Bauer method). The results showed that mouthwashes containing 0.5% chitosan solution and 25% red dragon fruit peel extract, 0.5% chitosan solution mouthwashes, and 25% red dragon fruit peel extract mouthwashes had moderate inhibitory activity (8, 12-9.96 mm) which significantly inhibited the growth of S. sanguinis bacteria and there were significant differences between each group. The study concluded that mouthwash containing 0.5% chitosan solution and 25% red dragon fruit peel extract (Hylocereus polyrhizus) had moderate effectiveness in inhibiting the growth of S. sanguinis bacteria.
