Potensi Beberapa Konsentrasi Ekstrak Daun Sirih Hijau (Piper betle L.) terhadap Streptococcus Mutans ATCC® 25175TM (In Vitro)
Abstract
Streptococcus mutans is the main pathogenic bacteria that causes caries. Green betel leaf medicinal plant (Piper betle L.) has antibacterial substances in the form of essential oils, phenols, kavikol, alkaloids, flavonoids, saponins, tannins, and steroids. The aim of the study was to determine the inhibition zone, Minimum Inhibitory Contentration (MIC) and Minimum Bactericida Contentration (MBC) of green betel leaf extract against Streptococcus mutans ATCC 25175TM concentrations of 20%, 40%, 60% and 80%. The method used was laboratory experimental with a post test only control group design research design. Determination of the inhibition zone using the Kirby Bauer standard blank disc diffusion method was measured using a digital caliper, while the KHM and KBM dilution methods were used to determine the level of turbidity and clarity. Green betel leaves were extracted using the maceration method, grouped into 4 treatments and 4 times replication. Data analysis used the Oneway Anova test to determine the significant difference for each concentration, followed by the Post Hoc Least Significance Difference (LSD) test to determine the significant differences in each concentration. The results showed that the average diameter of the inhibition zone at concentrations of 20%, 40%, 60% and 80% was 11.93mm, 13.12mm, 14.75mm and 17.19mm. Value of Minimum Inhibitory Contentration (MIC) at a concentration of 20%. Minimum Bactericida Contentration (MBC) at a concentration of 60%. The conclusion is that there is a significant potential for green betel leaf extract on the growth of Streptococcus mutans ATCC® 25175TM with a significance value (p=0.000), the diameter of the strongest inhibition zone is 80% concentration, the average diameter is 17.19mm, the higher the concentration, the potency of the extract will increase
