    Preparasi, Ekstraksi dan Analisis Kafein pada Urin Pengonsumsi Kopi (Coffea robusta)

    Abstract
    Caffeine (C8H10N4O2) is a natural stimulant that is quickly absorbed by the body. The FDA-approved caffeine dose is 100-200 mg/day. Caffeine has the effect of increasing heart rate, gastric secretion, and basal metabolism causing changes in blood pressure, increased diuresis relaxes smooth muscles. Excess consumption causes restlessness, anxiety, agitation, muscle tremors, sensory disturbances, heart palpitations, nausea, and vomiting diarrhea, even in certain cases can cause death due to overdose of stimulant drugs. This study aims to analyze and determine the levels of caffeine contained in the urine of coffee consumers (Coffea robusta). Qualitative analysis obtained by the parry method produces a dark blue or green colored solution and thin layer chromatography produces an Rf value of 0.489. Quantitative analysis using UV spectroscopy with a wavelength 275 nm. Validation analysis for accuracy obtained a recovery value of 104.5%. Precision SD value: 0.0003225 and RSD: 0.032 %. Linearity obtained the equation y = 0.0001x + 0.1646 and R2 = 0.9991, LOD 0.0059 mg/L and LOQ : 0,732 mg/L. Based on the analysis results obtained from the urine of coffee drinkers that have been developed and produce valid analytical data. Characterization using FTIR shows nicotine in urine samples of coffee drinkers as evidenced by the presence of clusters C-N (aliphatic amine), HC=O,C=H (Aldehyde), C = O (ketone), C = C (aromatic), C-H aromatic and C-H group (aliphatic). isis obtained from coffee drinkers that has been developed and produces valid analytical data.
