Isolasi dan Identifikasi Golongan Senyawa Flavonoida dari Daun Tumbuhan Kenitu (Chrysophyllum Cainito L)
Abstract
The flavonoid compound has been isolated from the leaves of Kenitu (Chrysophyllum cainito L.). Isolation was carried out by maceration extraction method with methanol solvent and the methanol extract was re-extracted with ethyl acetate repeatedly until it was negative for 5% FeCl3. The ethyl acetate extract was concentrated and redissolved with methanol then partitioned with n-hexane until the n-hexane layer was clear. The concentrated methanol extract was analyzed by thin layer chromatography and separated by column chromatography with silica gel 60 F254 as the stationary phase and chloroform : methanol as the mobile phase (90:10; 80:20; 70:30; 60:40) v/v respectively. The compounds obtained from the 10-60 fraction were purified by preparative thin layer chromatography using chloroform: methanol (90:10) v/v to produce 8 mg of green amorphous solid with a value of Rf 0.52 using chloroform: methanol (90:10) as eluent. ) v/v. Analysis of the compounds obtained was carried out using a UV-Vis, FTIR, and 1H-NMR spectrophotometer. The UV-Vis spectrum shows a wavelength (λ max) of 405 nm in the band I and 224 nm in band II . The FT-IR spectrum shows the presence of OH stretching, C-H stretching, C=O stretching, C=C stretching, and C-H bending, C-O stretching, C-O-C stretching groups. successively at wave numbers 3423.65 cm-1, 2852.72 cm-1, 1726.29 cm-1, 1512.19 cm-1, 1462.04 cm-1, 1249.87 cm-1, 1103, 28cm-1. The 1H-NMR spectrum shows the presence of protons H-2’, H-5’,H-6’, H-6, OH and OCH3. Based on the results of analysis and interpretation of the data, then the resulting compound The isolation is a flavonoid compound of the flavonol group.
