dc.description.abstractThe Sumatra fish, Puntius tetrazona, is an ornamental fish native to Indonesia with an attractive appearance. This research aimed to analyze the karyotype of Sumatra fish chromosomes, including the number, the size, and the shape. The descriptive method was used in this study, with the application of 0.007% colchicine for 12 hours. Chromosome preparation was carried out using the solid tissue technique, and staining was performed with 5% Giemsa stain. Data analysis was conducted using IdeoKar 1.3 software, and the karyotype of the chromosomes was constructed using Adobe Photoshop CS 3. The results of the study showed that the Sumatra fish had total 25 chromosomes diploid with sizes ranging from 1.16 µm to 5.35 µm, and the largest value of r was 0.93. The largest chromosome had a relative length percentage of 5.57 and a chromosome shape percentage of 2.54. The highest centromere index obtained was 0.47. Furthermore, the study found a TF% value of 40.48%, an Ask% value of 59.51%, an XCI value of 0.38, and an HCL value of 90.08. This research provided an understanding of the karyotype characteristics of Sumatra ornamental fish and had potential in genetic studies of fish.en_US
