dc.description.abstractBackground. Noise is one of the factors of stress and often disturbing around our neighborhood. Exposure to high noise more than 85 dB and continuously can cause permanent damage Organ of Corti and the auditory nerve. The Purpose. Assessing the outer hair cell damage in the Organ of Corti with noisy intensity by assessing the increase in the expression of Hsp 70 to do with the increase in p53, Cytochrome C and Caspase 3 as executor of apoptosis that causes permanent damage to the cochlear tissue Research Methods. Vivo experimental method using Wistar rats (Rattus norvegicus) with 5 treatment groups, namely PO (control), treatment group P1 (30- 50 dB), P2 (60-70 dB), P3 (80-90 dB) and P4 (100- 110 dB). Examination phase I assess the initial parameters of the outer hair cell damage Organ of Corti with Otoacustic of emissions (OAE), the assessment notice results SNR difference before and after treatment of more than 85 dB noise. Examination Phase II to assess the expression of Hsp 70 immunohistochemistry with p53, Cytochrome C and Caspase 3 as executor of apoptosis. Examination stage Ill by assessing cell ultrastructure Organ of Corti phase Ill assesses the percentage of outer hair cell ultrastructure damage to Organ of Corti The Results. Phase I results showed OAE noisy intensity of more than 85 dB of impairment SNR> 6 the damage to the outer hair cells and supporting cells in the Organ of Corti of the cochlea strated day 2th and day 8th and day. Phase II results in immunohistochemistry. Very strong relationship between Hsp 70 vs p53 (r = 0.886), Hsp 70 vs. Cytochrome C ( r = 0.919), Hsp 70 vs. Caspase 3 r = 0.925, Hsp 70 vs. Intensity noise r = 0943, p53 vs Damage Cochlear (r = 0.725) Cytochrome C vs. Caspase 3 .r = (0847), Cytochrome C vs. intensity noise (r = 0873), Cytochrome C vs Cochlear damage (r = 0683) Caspase 3 vs. intensity noise( r = 0932) Caspase 3 vs Cochlear damage . (r = 0.663) Cochlear damage i vs intensity noise (r = 0756) All of biomaker apoptosis was significantly different p < 0.01 all groups. Phase Ill results of Scanning Electron Microscope (SEM) in the Organ of Corti in the cochlea PO group (0- noise intensity 25 dB) the absence of damage to the cochlea, the group P1 (30-50 dB noise intensity) and a group P2 (60-70 dB noise intensity ) <50% damage organ of Corti, the group P3 (intensity 80-90) dB and group P4 (100-110 dB noise intensity) has been damage to more than 50% in the organ of corti of the cochlea. Discussion. The research hypothesis is proven by value; Caspase 3 -2140+ 2,260. intensitas noisy. The strong association between caspase 3 and sgnifikan noisy intensity due to the intensity of noise above 85 dB and trigger the heat shock response continually and will cause cell death organ of corti of the cochleaen_US
