dc.description.abstract | Natural phenolic compound is mostly obtained from solanum species and it plays an important fole in antimicrobial and antibacterial medication. Solanaceae family has secondary metabolite content in the form of phenolic, flavonoid, and tannin compounds and it may contain antibacterial activity. Lancing leaves belong to Solanaceae that contains chemical compunds sucah as alkaloids, flavonoids, tannins, saponins, steroids/triterpenoids, and glycosides, which also have potential as antibacterial. The research aims to find out the characterization simplicia, phytochemical screening, antibacterial activity of thanol extracts and lancing leaves (Solanum mauritianum Scop.) fraction as well as test the total phenolic and flavonoid contents, and determine the chemical compound using LC-HRMS method. The characterization simplicia, including organoleptic, macroscopic, microscopic examinations, and determination of water concentration, is also conducted on ethanol extracts. The process extraction is carried out through maceration process using ethanol solvent then it is fractionated using n-hexane and ethylacetate solvents. Phytochemical screening is carried out on simplicia powder, extracts and lancing leaf fractions. The test of antibacterial activity toward each fraction is conducted using agar diffusion method by observing the inhibition zones of several test bacteria: Staphylococcus aureus ATCC 25923 and Pseudomonas aureginosa ATCC 19615, as well as testing the concentration level of total phenols, total flavonoid and determining the chemical compounds through LC-HRMS. Based on the results of simplicia characterization, it is found thath water concentration is at 5.94%. The phytochemical screening test of ethanol extract indicates alkaloids, flavonoids, glycosides, saponins, tannins, steroids/ triterpenoids contents. The ethyl acetate fraction contains the highest antibacterial activity of Staphylococcus aureus at a concentration of 500 mg/ml with an effective inhibition diameter of 25.60 mm, and Pseudomonas aureginosa is at 500 mg/ml for 20.43 mm. The fraction showing the best antibacterial test is the ethyl acetate fraction. The test result of total phenolic content on ethyl acetate fraction of lancing leaves is 103.7648 mg GAE/g sample and the total flavonoid content is 16.6448 mg QE/g sample. Based on LC-HRMS Chemical Compound Test of ethyl acetate fraction, chemical compounds of Cianidanol, Gallic acid, Kaemprefol, Quercetin, Astragalin are found. | en_US |
