Wound is a condition where the network unit (skin) is lost or broken due to injury which generally disrupts normal cellular processes. Incisions or open cuts are wounds that occur because of being cut by a sharp instrument. Use of Castanopsis costata (Blume) A.DC leaves. in the karo community as a wound medicine has been around for a long time, it's just that the basis for using plants as medicine and their efficacy is known based on experience from generation to generation, without scientific research. The purpose of this study was to evaluate the extract ointment and Costanopsis costata leaf fraction ointment in accordance with the requirements of ointment preparations, to evaluate the wound healing activity of open incisions from the extract ointment and Costanopsis costata leaf fraction ointment preparations on male white mice and to find out the main compounds in the ethanol extract Costanopsis costata leaves were analyzed using GCMS. This research was carried out experimentally which included the manufacture of Costanopsis costata leaf simplicia, manufacture of Costanopsis costata leaf extract and fractions, preparation and evaluation of ointment preparations of Costanopsis costata extract and leaf fractions. Ointment preparations were tested for their healing activity in open incisions (incisions) and histopathological tests were carried out. Furthermore, the ethanol extract of Costanopsis costata leaves was analyzed using GCMS to determine the main compounds. The results showed that the extract ointment and the leaf fraction ointment of Costanopsis costata fulfilled the evaluation requirements of ointment preparations which included organoleptic tests, homogeneity and pH tests. Extract ointment and leaf fraction ointment of Costanopsis costata also showed healing effect on cuts. EEDCC ointment provides a wound healing effect at a concentration of 30% and 45%, 30% FADCC ointment, 15% to 45% FEADCC ointment provides a wound healing effect, while FnHDCC ointment at a concentration of 30%. The GCMS EEDCC results contain the main compounds, namely Methyl Linolenate and Phytol Isomers. Histopathological test results showed that the EEDCC ointment showed a higher number of fibroblass and dense collagen compared to the other groups From the results of the study it can be concluded that the ointment preparations of Costanopsis costata extract and leaf fraction meet the evaluation requirements for ointment preparations, the ointment extract preparations and Costanopsis costata leaf fraction can have a healing effect on open incision wounds and the main compounds in Costanopsis costata leaf extract are Methyl Linolenate and Phytol Isomers.en_US
