Introduction: Periodontal disease is a local inflammatory disorder in which periodontal pathogenic bacteria can escape the host's immunological defense system that attacks the supporting tissues of the teeth so that it can cause tissue damage and bone loss. Scaling-root planing is the basic mechanical treatment to remove local deposits such as plaque, calculus, endotoxin and other local factors in periodontitis and has been shown to reduce pocket depth and increase the level of attachment. Tetracycline is a class of antibiotics in the treatment of periodontal disease and has been chemically modified as antimicrobial, anticollagenase and anti-inflammatory drugs that can reduce clinical parameters and increase the healing factor of PDGF- BB. Chitosan nanoparticles can be a drug delivery and have the properties of slower and controlled drug release, better drug solubility and stability. Purpose: This study aims to see the improvement of clinical parameters and PDGF-BB expression which were reviewed by immunohistochemistry. Materials and Methods: The research sample consisted of 33 male wistar rats which were divided into seven treatment groups. The control group, giving tetracycline 0.7% once and every day for 3 and 8 days, giving placebo gel once for 3 and 8 days. Clinical parameters and PDGF-BB expression were examined on the 0, 3 and 8 days after treatment with different mice. Results: The results of this study showed that changes in clinical parameters were better with daily tetracycline administration for 8 days and PDGF-BB expression was stronger on day 3 after treatment with statistically significant results (p < 0.05). Conclusion: Scaling with the application of chitosan nanoparticles loaded by tetracycline 0.7% every day for 8 days gave better changes in clinical parameters and increased PDGF-BB expression on day 3.
