dc.description.abstractSoft Denture Liner (SDL) is a soft material used as a soft coating to support the tissue of malfitting dentures. This material is used to resurface the denture, resulting in an accurate fit to the denture base. Most available SDL materials have several disadvantages, including porosity and Candida albicans colonisation. Prolonged use has shown that the use of SDL can facilitate the formation of Candida biofilms and poor adhesion to the RAPP base, leading to oral candidiasis. The addition of oregano (Origanum vulgare) essential oil gel has shown fungistatic and fungicidal properties against Candida albicans growth. The aim of this study was to determine the effect of adding oregano (Origanum vulgare) essential oil gel at concentrations of 3.12% and 6.25% to auto-polymerized silicone soft denture liners on the inhibition zone of Candida albicans during 7 and 14 days of use. This study used 28 specimens (n=28), namely cylindrical auto-polymerized silicone SDL with a diameter of 6 mm and a thickness of 1 mm. The treatment groups in this study were 4 treatment groups, namely the no addition group, the nystatin addition group, the 3.12% oregano essential oil gel addition group and the 6.25% oregano essential oil gel addition group. All samples were incubated at 37°C for 7 and 14 days. The data were analysed using univariate test to determine the mean and standard deviation of the inhibition zone of Candida albicans in each group. Oneway ANOVA test and Dunnett T3 test were used to determine the effect of the addition of oregano essential oil gel. The results of the univariate test analysis showed that the mean value and standard deviation of the inhibition zone of Candida albicans in the group without treatment with a usage time of 7 days was 0 mm, the addition of nystatin with a usage time of 7 days was 9.5 ± 0.44 mm, the addition of oregano essential oil gel 3.12% with a usage time of 7 days was 12.6 ± 0.85 mm and the addition of oregano essential oil gel 6.25% with a usage time of 7 days was 15.67 ± 0.55 mm. The no treatment group with a 14-day application period was 0 mm, the nystatin addition with a 14-day application period was 9.06 ± 0.31 mm, the oregano essential oil gel 3.12% addition with a 14-day application period was 11.54 ± 0.94 mm and the oregano essential oil gel 6.25% addition with a 14-day application period was 15.23 ± 0.62 mm. The results of the Oneway ANOVA test showed a significant effect of the addition of oregano essential oil gel (Origanum vulgare) 3.12% and 6.25% based on the application time of 7 days with p = 0.001 (p < 0.05) and 14 days with p = 0.001 (p < 0.05). Based on the results of this study, it can be concluded that the addition of Oregano (Origanum vulgare) essential oil gel at a concentration of 6.25% in an auto-polymerized silicone soft denture liner is recommended because it is able to inhibit the growth of Candida albicans up to 14 days of use compared to the addition of nystatin, although the inhibition zone decreases with the time of use.en_US
