Salmonella typhimurium is an intracellular bacteria that can affect the immune system so that an immunomodulator is needed to modulate the function and activity of the immune system. CD4+ and IL2 T cells are one of the parameters of the immune system that plays a role in salmonella infection. The use of medicinal plants as a traditional medicinal herb is an alternative in getting the effect of immunity with minimal toxicity. One of the herbs that play a role in immunity is pirdot leaves(Saurauia vulcani Korth ) which contains flavonoids that play a role in the inflammatory process. This study aims to determine the immunomodulatory effects of ethanol extract of pyrdot leaves (Saurauia vulcani Korth ) on CD4+ T cell profile and serum IL 2 levels from rat liver induced by Salmonella thyphimurium. Methods: experimental research with post-test only control group design. Male wistar rats are divided into 4 groups: I. Normal (KO), II. Negative (Kneg), III. Imboost 22.5 mg / kgBB(KPos), IV. Ethanol extract of pirdot leaves 500mg / kgBB (P). Group II-IV received an injection of S. thypimurium on Day 4. At the end of the treatment of animals tried to be sacrificed, blood was taken from the heart and excised liver organs for examination of IL 2 levels, histopathology and liver CD4+ T cells immunohistochemically. The data were analyzed by Kruskal-Wallis test followed by Whitney Man Test. The results of this study showed the highest levels of IL 2 in the group of pirdot leaf ethanol extract (2.23 ± 0.18),followed by KPos (1.86 ± 0.33),K0 (1.71 ± 0.40), and the lowest KNeg (1.64 ± 0.28).The average value of histopathological structure score in Kruskal-Wallis test showed significant differences in hydropic necrosis degeneration, inflammation, granuloma, port triad damage (p<0.05) while for bleeding there was no difference with a value of p=1.00. Comparison of the results of CD4 + T cell IRS scores there were significant differences in all groups with a value of p=0.004. Conclusion: ethanol extract of pirdot leaves at a dose of 500mg / KgBB increased CD4 + T cell IRS scores and serum IL 2 levels compared to other groups.en_US
