Cases of death of methamphetamine abusers in Indonesia were increasing,
where bodies are generally found after several days of investigation, so forensic
entomotoxicology uses insects to detect methamphetamine. The use of insects was
chosen because insects are the first living things to infest corpses. This study aims to
analyze methamphetamine found in necrophagous insect maggot grown on beef and
pork using GCMS as an application of forensic entomotoxicology and to determine
the validation method of the GCMS used. The methamphetamine contained in maggot
was extracted by maceration and sonication methods using methanol:chloroform 1:3
and then a preliminary test was carried out using the Marquis reagent which changed
the color of the sample to brownish orange. In the qualitative test, methamphetamine
was obtained from day 1 to 8 for in respectively : 26.00, 42.50, 30.53, 49.57, 47.78
μg/mL and in pork respectively: 16.95 , 30.92, 27.94, 434.55, and 34.23 μg/mL. The
TLC test produced an Rf value close to the comparator Rf value (0.648) which
indicated that maggot was positive for methamphetamine. The validation of the GCMS
method for methamphetamine compounds shows an accuracy value with a percent
recovery (% recovery) of 101,896 % with a straight line equation y = 190269.56x –
970.296 with a correlation value (R) of 0.991. The precision test value with a standard
deviation of 0.420 and a Relative Standard Deviation (RSD) of 1,186 % is <20%. The
LOD and LOQ values were 6.6222 x 10-6 μg/mL and 1.2514 x 10-4 μg/mL
respectively.MS.
