    Derajat Kerusakan Ginjal Akibat Pelvic Ureteral Junction Obstruction Buatan terhadap Kadar NGAL, IL-18, dan Gambaran Histopatologis pada Tikus Wistar

    Abstract
    Introduction Markers such as NGAL, and IL-8 have been associated with the degree of kidney injury that occurs due to an obstruction. This study aims to assess the degree of kidney injury due to pelvic ureteral junction obstruction on the levels of NGAL, IL-8, and histopathological features in wistar rats. Method This research is an experimental study on rats (Laboratory Animal Experimental Design). This research was conducted at the Pharmacology Laboratory of the USU Medical Faculty, the USU Medical Faculty Integrated Laboratory, and the Anatomical Pathology department of the USU Medical Faculty in November 2021 – December 2021. This study used 32 male wistar rats which were divided into three groups, namely negative control with a total of 4 rats, the group sham operation with a total of 4 rats, and the group that underwent ureteral ligation with a total of 24 rats. On day 0, rats from control and sham group were taken randomly and urine samples were taken for examination of NGAL and IL-8. Both rats will be decapitated and performed nephrectomy. The rat kidney was sent to the Anatomical Pathology department for histological examination. This process was repeated on hari 4, 7, 14, and 21. Results There was no significant difference in NGAL values between the intervention groups (p = 0.063). There was a significant difference in IL-18 levels between the intervention groups (p = 0.031). Significant differences in IL-18 were found between the control group and the PUJO group on day 4 (p = 0.028), the sham surgery group and the PUJO group on day 4 (p = 0.014), the PUJO group on day 4 and day 7 ( p = 0.008), and the 7th day PUJO group with 14th day (p = 0.033). Conclusion IL-18 levels can be used as a marker of kidney injury due to obstruction of the Pelvic Ureteral Junction.
