Background: Physical exercise is a process of increasing contraction of skeletal muscles involving several organ systems to increase energy formation and it is one of the pillars for the management of type 2 diabetes mellitus (TDM2). Such activity affects the cardiovascular, respiratory, endocrine and skeletal muscle systems. T2DM affects the expression of the TLR-4 gene, TGF-β, and renal mononuclear cells. Aims : 1) Analyze the effect of moderate continuous training and slow interval training in expression of TLR-4 gene, TGF-β and renal mononuclear cell activity in wistar rats T2DM model. Methods: A moderate intensity continuous training physical activity given to the diabetic rat group in the form of running on a treadmill with a running speed of 20 m/min for 30 minutes while the slow type interval training physical activity given to the diabetic rat group was in the form of running on a treadmill with a running speed of 20 m/minute for 10 repetition sessions, with a duration of 2 minutes per session with interspersed active breaks for 1 minute. TLR-4 gene expression was measured using the realtime R-PCR method. TGF-β gene expression was measured using real-time methods. Kidney mononuclear cells were measured by white blood cell count using image analysis software. Results: The results showed that in the continuous group, TLR-4 and monocytes decreased while lymphocytes increased after 8 weeks of research treatment and statistically obtained p > 0.05, but significant results were found on the decrease in TGF-β in the continuous group and statistically obtained the result p < 0.05. Likewise, in the interval group, TLR-4 and monocytes decreased while lymphocytes increased and statistically obtained a p > 0.05, but significant results were found against a decrease in TGF- β in the interval group and statistically obtained a p < 0.05. Moderate intensity continuous training was better at lowering the expression of the TLR-4 and TGF- β genes while slow-type training intervals were better at lowering kidney monocyte cells and increasing renal lymphocytes compared to moderate intensity continuous training in mice of the wistar strain of T2DM model. Conclusion: From this research, it can be concluded that there is an effect of continuous training of moderate intensity and slow-type training intervals on the expression of the TLR-4 gene, TGF-β and kidney mononuclear cells of wistar rats T2DM model.en_US
