dc.description.abstractIntroduction: In orthodontic treatment, tooth movement is always accompanied by inflammation. The acute physiological inflammatory process that the patient experiences ranges from discomfort to pain. Patients experience discomfort during all orthodontic procedures. The hydroxyl group in position 3 of the pyridine structure of vitamin B6 has biological activity, functioning as a potent antioxidant and anti-inflammatory molecule. By enhancing the pain threshold, vitamin B6 can serve as an analgesic. This study seeks to investigate the effect of vitamin B6 administration on reducing inflammation-induced pain in Wistar rats undergoing orthodontic tooth movement. Methods: Samples of Wistar rats were divided into four categories. One group received no supplements and orthodontic force. One group was administered no supplement and orthodontic force for 0, 6 hours, 1, 3, and 7 days. One group was administered paracetamol and orthodontic force for 0, 6 hours, 1, 3, and 7 days. The remaining group received vitamin B6 supplements and orthodontic force for 0, 6 hours, 1, 3, and 7 days, respectively. Using macro photometry, linear distance measurements were taken and analyzed using the Corel Draw application. Each group's GCF was extracted from Wistar rats using paper points, centrifuged, and subjected to an ELISA test to determine the SP levels. Results: The MANOVA test revealed significant differences between the control, paracetamol, and vitamin B6 groups regarding linear distance (p = 0.000< 0.05). However, there was no statistically significant difference between the control group and vitamin B6 at each treatment time difference. The highest average linear distance occurred seven days after given orthodontic force. The MANOVA test on SP levels revealed significant differences between the paracetamol and vitamin B6 groups based on the time interval (p = 0.042< 0.05). In the vitamin B6 treatment group, the Pearson test revealed that the relationship between SP and linear distance was positive but weak, with a value of r = 0.128. Conclusion : Vitamin B6 supplementation is ineffective in reducing pain (SP) during orthodontic tooth movement.en_US
