The number of elderly people increases every year. Increasing age in the elderly is followed by frailty which causes changes in the body's physiology and ultimately can result in the emergence of various diseases that can develop into chronic ones. Efforts need to be made to prevent the occurrence of chronic diseases in the elderly and elderly, through early examination efforts to prevent this. One of the genes that acts as a biomarker for frailty is the SIRT1 gene. This study aims to determine the relationship between the rs7895833 polymorphism in the SIRT1 gene and plasma SIRT1 levels and frailty in elderly people. This research was a cross-sectional type with pre-elderly and elderly patients at the North Sumatra University Hospital with 132 research subjects having their blood taken and interviews conducted to determine the degree of frailty. The blood of the research subjects was examined for SIRT1 plasma levels using ELISA and the rs7895833 polymorphism. The research results showed that of the 132 research subjects, 51.5% were in the strong category, 32.6% pre-weak, and 15.9% weak. Meanwhile, when measuring SIRT1 plasma levels, the mean SIRT1 plasma levels were 56.11 ± 31.51. The results of the SIRT1 rs7895833 gene polymorphism showed AG (56.8%), GG genotype (31.1%) and AA (12.1%). There was no relationship between plasma SIRT1 levels and the SIRT1 rs7895833 gene polymorphism (p = 0.983) and SIRT1 levels with the degree of weakness (p = 0.788), and there was no relationship between the SIRT1 rs7895833 genotype and the degree of weakness (p = 0.084). This research proves that there is no relationship between the SIRT1 rs7069102 gene polymorphism and the degree of weakness in pre-elderly and elderly patients at North Sumatra University Hospital.
