Background. Frailty is defined as a syndrome of age-related physical decline characterized by significant vulnerability to health issues. The diagnosis of frailty is established based on three or more of five criteria, which include weakness, reduced walking speed, rapid fatigue, decreased physical activity, and weight loss. Frailty is not a disease but rather a state between functional and non-functional, and between health and illness. Sirtuin 1 is known to play a role in frailty. In solder individuals, an improvement in nutritional status is positively associated with the expression of the sirtuin 1 gene regardless of frailty status. However, research on this topic is still very limited. Therefore, studying the relationship between the polymorphism (rs2273773) in the sirtuin 1 gene, plasma sirtuin 1 levels, and frailty in the elderly is of utmost importance. Methods. This study employed a cross-sectional method involving the observation or measurement of research subjects at a single point in time to determine the relationship between the polymorphism (rs2273773) in the sirtuin 1 gene, plasma sirtuin 1 levels, and frailty in the elderly. Subjects were selected using non-probability sampling, with a total of 118 samples. Results. Pearson correlation analysis showed a significant relationship between the polymorphism genotype (rs2273773) in the SIRT1 gene and the frailty scale (p-value < 0.05), while no significant relationship was found between plasma SIRT1 levels and the frailty scale (p-value > 0.05). Furthermore, the F-test indicated a significant simultaneous relationship between the polymorphism genotype (rs2273773) and plasma SIRT1 levels with frailty (p-value < 0.05). Conclusion. There is a relationship between the polymorphism of the SIRT1 gene (rs2273773) and plasma sirtuin 1 levels with frailty in the elderly.en_US
