    Hubungan Polimorfisme RS7069102 pada Gen SIRT 1 dengan Kadar SIRT 1 Plasma dan Kelemahan(FRAILTY) pada Orang Lanjut Usia

    Abstract
    The number of elderly people is increasing every year. Aging in the elderly will be followed by frailty which can result in cumulative changes, ultimately resulting in chronic disease. To prevent the occurrence of chronic disease in the elderly, early examination is needed to prevent this. The SIRT1 gene has potential as a biomarker for frailty. This study aims to determine the relationship between the rs7069102 polymorphism in the SIRT1 gene and plasma SIRT1 levels and frailty in elderly people. This research was a cross-sectional study with elderly patients at North Sumatra University Hospital as subjects. A total of 125 research subjects had their blood taken and interviews were conducted to determine the weakness index. The blood of the research subjects was examined for SIRT1 plasma levels using ELISA and the rs7069102 polymorphism. The research results showed that of the 125 research subjects, 52.8% were in the strong category, 32.8% pre-weak, and 14.4% weak. The results of measuring SIRT1 plasma levels showed that the average SIRT1 plasma level was 56.47 ± 31.35. The SIRT1 rs7069102 gene polymorphism was found to be CG (73.6%), CC genotype (22.4%) and GG (4%). A non-significant relationship was found between plasma SIRT1 levels and the SIRT1 gene polymorphism rs7069102 (p = 0.337) and SIRT1 levels and the degree of weakness (p = 0.888). There was a relationship between the SIRT1 rs7069102 genotype and the degree of weakness (p= 0.008). This research proves that there is a relationship between the SIRT1 rs7069102 gene polymorphism and the degree of weakness in elderly patients at North Sumatra University Hospital.
