dc.description.abstract | Turmeric (Curcuma domestica Val.) is one of plants growing in Indonesia
that has numerous health advantages. Curcuminoid compounds are turmeric
contents that are known to have immunomodulatory activity and have been proven
in several in vivo test. Immunosupressive medication, such as cyclophosphamide,
is widely used to treat cancer, which can cause susceptibility to infection. Therefore,
the aim of this study is to determine the immunostimulant effect of curcuminoid
VCO extract from turmeric rhizomes on white mice that have been infected with
Staphylococcus aureus and induced by cyclophosphamide.
Curcuminoid VCO extract is obtained using the Microwave Assisted
Extraction (MAE) method with VCO as a solvent. Then quantitative analysis of the
VCO curcuminoid extract is carried out using TLC-Densitometry. The
experimental animals used are male mice which are given VCO curcuminoid
extract at a dose of 1 mL, 0.5 mL, 0.25 mL, curcuminoid suspension 200 mg/kgBW
1 mL, levamisole suspension 25 mg/kgBW 1 mL, VCO 1 mL, and suspension
CMC-Na 0.5% 1 mL as a negative control on days 1 to 14. On the 4th day, the
experimental animals are infected with Staphylococcus aureus bacteria, then on
days 4, 8, and 12 they are induced by cyclophosphamide 30 mg/kgBW, furthermore
the immunostimulant effect is tested using the total leukocyte and differential
leukocyte analysis method using a hematology analyzer, IgG and IgM antibody
levels, levels of cytokines IL-2, IL-4, IFN- ℽ, and the amount of TLR4 using the
ELISA method and microscopic examination of mice spleen pulp Hematoxylin Eosin preparations.
The test results show that curcuminoid VCO extract 1 mL, 0.5 mL, 0.25 mL,
curcuminoid suspension, levamisole suspension, and VCO are able to increase total
leukocytes, leukocyte differential, antibody levels, cytokine levels, TLR4 levels,
and decrease pulp diameter size of mice spleen with a significant difference (p <
0.05) to the CMC-Na 0,5% group as a negative control.
Based on the results of this study, VCO curcuminoid extract can increase the
number of total leukocyte and leukocyte differential, levels of cytokine IL-2, IL-4,
and IFN-ℽ, TLR4 levels, and decrease spleen pulp diameter most effectively at a
dose of 1 mL of curcuminoid VCO extract. Therefore, curcuminoid VCO extract
has the potential to have an effect as an immunostimulant. | en_US |
