This research was conducted at the Plant Tissue Culture Laboratory, Faculty of Agriculture and Phytochemistry Laboratory, Faculty of Pharmacy, University of North Sumatra from February 2023 to July 2023. The Complete Randomized Design (RAL) was used with 2 treatment factors and 3 replicationss. The first factor was the concentration of PGR in MS (K) media: K0 (NAA 1 ppm + kinetin 0 ppm), K1 (NAA 0.5 ppm + kinetin 0.5ppm), K2 (NAA 1 ppm + kinetin 1 ppm).
The second factor was the concentration of Salicylic Acid (M): M0 (Salicylic Acid 0 ppm), M1 (Salicylic Acid 0.5 ppm), M2 (Salicylic Acid 1 ppm). The parameters observed were callus morphology, SCV (Settled Cell Volume), PCV (Peacked Cell Volume) and analysis of secondary metabolite content. The results showed that the combination of PGR NAA and kinetin and the combination of Salicylic Acid significantly affected all parameters. The concentration of K1M1, K2M1, K2M2 showed optimal growth with an average of 0.03 grams.
