Objective: This study was conducted to analyze the comparison of peritumoral budding appearance in colorectal adenocarcinoma NOS stained with hematoxylin-eosin (HE) and pancytokeratin (AE/AE3) immunohistochemistry.
Methods: This study was an analytic cross-sectional, using 48 samples of paraffin block cases of colorectal adenocarcinoma NOS that had been diagnosed at the Anatomical Pathology Laboratory of H. Adam Malik General Hospital Medan and at the Anatomical Pathology Laboratory, Faculty of Medicine, University of Sumatera Utara. Peritumoral budding was counted on preparations stained with Hematoxylin-Eosin and pancytokeratin (AE1/AE3) using an Olympus CX 43 light microscope, 20 mm ocular diameter. Select "hotspot" by viewing all slides at 10x magnification, after selecting "hotspot" calculate the initial budding with 20x magnification on a field area of 0.785 mm. Reporting of tumor budding uses a three-level scoring system, namely: 0–4 buds: low budding, 5–9 buds: intermediate budding, ≥10 buds: high budding. Data were analyzed using Pearson's correlation test and Friedman's test.
Results: There is a statistically significant relationship between peritumoral budding appearance and HE staining and pancytokeratin (AE1/AE3) immunohistochemistry in colorectal adenocarcinoma NOS. It can be concluded that there is no difference in assessing peritumoral budding with HE and pancytokeratin (AE1/AE3) staining.
Conclusion: Peritumoral budding assessment is suggested by HE staining.
