dc.description.abstractBackground : Currently, obesity is a global health problem due to the high mortality rate of disease complications caused by obesity. Losing weight means reducing complications, by inhibiting the processes and pathways involved in the process of adipogenesis. Objective : To prove whether binahong leaf extract (Anredera cordifolia) can reduce the body weight by measuring levels of ERK, PI3K and PPARγ expression in white Wistar rats of the obesity model. Method : This study is a pure experiment with randomized post test only control group design, 36 Wistar rats were divided into 6 groups, namely with 3 control groups namely K1 (standard diet without treatment), groups given a high-fat diet, K2 (without treatment), K3 (orlistat) and 3 treatment groups given a high-fat diet and binahong leaf extract at various doses namely P1 (dose 50 mg / kgBW), P2 (dose 100 mg / kgBW), P3 (dose 150 mg / kgBW). Measurement of body weight (BW), body length and abdominal circumference (AC) before and after giving diet and treatment. Determination of obesity >20% body weight gain after dieting. Examination of ERK and PI3K levels by ELISA method and immunohistochemical examination of PPAR γ expression in abdominal fat adipose tissue of rats. Data analysis with One way Anova test on normally distributed samples and continued with LSD post-hoc test, if p<0.05, abnormal sample distribution Kruskal-Wallis test, followed by Mann Whitney test, if p<0.05. To see the difference before and after induction and treatment in each group, dependent T test was conducted and Wilcoxon test. Data were significant when p<0.05. Results : There was a decrease in BW in the administration of Anredera cordifolia ethanol extract at all doses (50 mg/kgBW,100 mg/kgBW and 150 mg/kgBW), statistically not significant (p>0.05), but there was a significant decrease in AC at doses (50 mg/kgBW, 100 mg/kgBW). There was a decrease in ERK levels at doses of 50 mg/kg BW and 100 mg / kg BW statistically not significant, there were significant differences in PI3K levels and PPAR γ expression in the control group and the treatment group (p <0.05), PI3K levels increased in the use of doses of 50 mg/kgBW, 100 mg / kg BW compared to the control group of high fat diet without treatment (P2) and low PPAR γ expression at all doses (50 mg/kgBW, 100 mg/kgBW and 150 mg / kgBW). Conclusion:Binahong leaf ethanol extract reduces body weight by affecting the adipogenesis process by suppressing PPARγ expression and ERK levels and increased PI3Ken_US
