Background : The incidence of head injuries in the world is currently touching 250 cases per 100,000 population worldwide. Two types of cell death can occur in head injury, namely necrosis and apoptosis. Polyphenols are natural chemical compounds as antioxidants that help protect cells from free radicals and can prevent the risk of various diseases so that researchers are interested in examining the effect of giving Oleuropein to rats with traumatic brain injury models using AIF, GFAP, MMP-9, and HMGB parameters -1. Objective : To prove the effect of administering Oleuropein isolate on the expression of Apoptosis Inducing Factor (AIF), Glial Acidic Fibrillary Protein (GAFP), Matrix Metalloproteinase-9 (MMP-9), and High Mobility Group Box-1 (HMGB-1) in rats with the traumatic brain injury model. Methods : This research is an experimental research, laboratory observation. Performed at the Biology Laboratory, Faculty of Medicine, University of Brawijaya Malang from June 1st – Desember 30th 2022. Examination of brain tissue preparations was carried out at the Department of Anatomical Pathology FK Universitas Brawijaya to prove the effect of Oleuropein isolate on the expression of AIF, GAFP, MMP-9, and HMGB-1 in rats with traumatic brain injury models. Oleuropein isolate obtained from Sigma-Aldrich, carried out by a rat treating expert at a dose of 50-100 mg/KgBW in rats Results : The results showed that GFAP expression increased significantly in the TBI group compared to the normal group (p < 0.05), in line with the expression of HMGB1 and MMP9 which was also higher in the TBI group compared to the normal group (p < 0.05). The results showed that the expression of GAFP (microglia cells) decreased significantly in the TBI group with OLE administration compared to the TBI group (p <0.05), and there was a significant decrease in HMGB1 and MMP-9 expression in the TBI group with OLE administration compared to the TBI group without OLE administration. (P < 0.05) Conclusion : There is an effect of giving Oleuropein isolate on the expression of Apoptosis Inducing Factor (AIF), Glial Acid Fibrillary Protein (GAFP), Matrix Metalloproteinase-9 (MMP-9), and High Mobility Group Box-1 (HMGB-1) in rats with traumatic brain injury rat models
