Antibacterial Effectiveness of Ethanol Extract of Red Grape Seed (Vitis vinifera L.) Against the Growth of Streptococcus mutans ATCC® 25175TM (In Vitro)
Abstract
Streptococcus mutans is the main microorganism that causes caries, Gram-positive bacteria, facultative anaerobes, coccus form, non-motile, chain-like arrangement, acidogenic and aciduric. The main caries treatment uses chlorhexidine gluconate 0.2% as an antiplaque agent. As an alternative, a substitute for the mouthwash was made from red grape seed extract (Vitis vinifera L). Red grape seeds contain phytochemical compounds flavonoids, tannins, terpenoids, and saponins. The purpose of this study was to determine the inhibition, Minimum inhibitory concentrations (MIC), minimum bactericidal concentration (MBC) at the most significant concentration against Streptococcus mutans ATCC® 25175TM. Experimental laboratory design method with post test only control group design. Determination of inhibition using disc diffusion method, while MIC and MBC by dilution method to determine the level of turbidity and clarity. Red grape seeds were extracted using maceration method, grouped into 7 treatment groups and 4 repetitions. Data analysis using Kruskal-Wallis test and ANOVA then continued with Mann-Whitney Test and Post Hoc Test. The result on the inhibition of ethanol extract of red grape seeds against the growth of Streptococcus mutans ATCC® 25175TM with the strongest concentration is 50% (23.1 + 1.16), MIC at a concentration of 20%, MBC 50%, and the most significant concentration is 50%. The conclucion is that ethanol extract of red grape seeds is effective in inhibiting the growth of Streptococcus mutans ATCC® 25175TM bacteria
