Stress in young adults has an impact on sleep quality which ultimately affects the hormone melatonin and also has an impact on the alpha-amylase enzyme as a stress biomarker. Drinking black tea can be an effort to increase relaxation because it contains theanine. However, to date there has been no research comparing the effect of drinking Sidamanik black tea on levels of the salivary hormone melatonin and salivary alphaamylase in young adults. The aim of this study was to analyze the influence of melatonin hormone levels in saliva and salivary alpha-amylase protein activity before and after drinking black tea in faculty of dentistry on university of Sumatera Utara students. This research method was pre-experimental which was carried out using a one group pretestposttest design approach. The population in this study was faculty of dentistry on university of Sumatera Utara students aged 18-25 years who were taken using purposive sampling with inclusion and exclusion criteria totaling 20 subjects. Saliva was collected before and 30 minutes after drinking Sidamanik black tea. Melatonin levels were measured using the ELISA method and alpha-amylase activity with a spectrophotometer. The results showed that there was no significant difference in the melatonin hormone (p=0.204) after drinking Sidamanik black tea. Meanwhile, there was no significant difference in alpha-amylase (p=0.005) after drinking Sidamanik black tea. The conclusion of this study is that there is an effect of Sidamanik black tea by increasing the hormones melatonin and alpha-amylase. The results of the study showed that there was no significant difference in the melatonin hormone (p=0.204) after drinking Sidamanik black tea.
