dc.description.abstractThe need for 70% food emulsifier was mono glycerin form. There was concern about monoglyceride products that were produce as unfriendly manner and most of the products are still import, even though Indonesia has the potential to become a monoglyceride producer due to the availability of raw materials such the form of vegetable oil (palm oil). This study aims to analyze the optimum conditions for the enzymatic glycerolysis process of palm oil derivatives using the TLIM lipase with parameter of substrate type, substrate ratio, temperature and time for MG production and to analyze the characteristic of TLIM in catalyzing the optimization process if enzymatic glycerolysis on the formation of MG with different type of palm oil derivatives, namely Refined Bleached Deodorized Palm Oil (RBDPO), Fully Hydrogenated Palm Oil (FHPO) and Fully Hydrogenated Palm Kernel Oil (FHPKO). The research design used was the Behnken Box Design (BBD) with parameter (highest-lowest) successively was temperature (50-90°C), triglyceride (TG): glycerol (Gly) substrate ratio (0.27-1 mol/ moles) and time (8-24 hours). The MG concentration was identified using the Gas Chromatography method. Furthermore, regression analysis and optimization of responses to research data obtained using the Minitab application were carried out. Analysis of the description of the optimization of the enzymatic glycerolysis process was carried out using the Response Surface Methodology (RSM). Analysis of the characteristics of the TLIM lipase was performed using a correlation test and a histogram. Regression analysis showed that the parameter of the optimization process that had a significant effect at quadratic and interaction were temperature and time (hours) x substrate ratio (TG.Gly) with the alpha value <0.05 obtained on RBDPO substrates, respectively. Optimum conditions were obtained on the enzymatic glycerolysis process of RBDPO substrate with the concentration of MG > 49% , TG:Gly ratio (1:1 mol/mol), temperature 69.85°C and 8 hour's reaction time. Analysis of the characteristics of TLIM lipase was influenced by different types of substrate (t test <0.05) with the best activity in catalyzation of the glycerolysis reaction for MG production obtained on RBDPO substrate. The best selectivity of TLIM lipase was obtained on TG substrates containing fatty acid with chain length of C 16 to 18 atoms with double bonds 1 and 2 on C 18 atoms. However, the TLIM selectivity was not good on TG substrates containing fatty acid chains of C 12 to 14 with saturated bonds. Suggestions for further research are to carry out the enzymatic glycerolysis reaction with a time of more than 24 hours on FHPO substrates to obtain greater levels of MG and it is necessary to carry out molecular distillation on the most optimal MG product in order to obtain higher levels of MG.en_US
