The phenolic compound had been isolated from the stem bark of Pucuk Merah (Syzygium myrtifolium Walp. Isolation was carried out by maceration extraction method with methanol and methanol extract was re-extracted with ethyl acetate repeatedly until negative to 5% FeCl, Ethyl acetate extract was concentrated and reconstituted with methanol and partitioned with n-hexane until the n-hexane laver was clear. The methanol extract was analyzed by thin layer chromatography and separated by column chromatography with a stationary phase of silica gel and a mobile phase of chloroform: methanol (90:10: 80:20: 70:30; 60:40) Fractions 86-160 was purified by preparative thin layer chromatography using chloroform: methanol (80:20), eluent to produce a greenish red amorphous solid of 7.3 mg at Rf value of 0.6 using chloroform methanol (80:20) %, as eluent and Rf 0,66 using acetone, benzene (60:40)%, eluent. Based on the analysis of the UV-Visible spectrophotometer, it was a wavelength (max) of 275 nm, the FT-IR spectram shows the presence of OH, C=C aromatic, C=O ester, C-H., and C-O groups. The proton Nucleus Magnetic Resonance Spectrum ('H-NMR) shows the presence of H-2. H-6 protons, proton from CH. bins to the benzene ring, and methoxy protons. Based on the data analysis and interpretation, the isolated compound was a phenolic compound derived from phenolic acid. It was found that total flavonoids from the stem bark of the Pucuk Merah plant were strong against gram-positive and negative bacteria. Evidenced by the clear zone produced against Stupylococcus aureus bacteria of 11.9 mm 75%; 13 mm 50%; 12.2 mm 25%; 11.7 mm 100% Likewise with the resulting clear zone against Escherichia coli bacteria of 12.5 mm 75% 13,1mm 50%: 14 man 25%; 13,2 mm 10%
