Topography and Antibacterial Effectiveness of Chitosan Chips Tetracycline 0.7 % in Inhibiting Periodontal Pathogen Bacteria
Abstract
Background: The main cause of periodontitis is periodontal pathogenic bacteria, such as Porphyromonas gingivalis, Aggregatibacter actinomycetemcomitans and Fusobacterium nucleatum. Local drug delivery for the treatment of periodontal disease has received great attention during the last decade due to the side effects of systemic antibiotics. One form of local drug delivery for periodontal treatment is periodontal chips. Tetracycline has been proven to inhibit the growth of periodontal pathogenic bacteria. Chitosan is a natural biopolymer that has potential properties as a drug carrier.
Objective: This research aims to determine the topography and effectiveness of chitosan chips 0.7% tetracycline as an antibacterial to inhibit the growth of periodontal pathogens.
Method: The chip topography was seen using SEM and FTIR was carried out to see the functional group bonds. Antibacterial effectiveness against the bacteria A. actinomycetemcomitans, P. gingivalis and F. nucleatum was carried out. Bacterial samples were obtained from pure cultures of P. gingivalis (ATCC33277), F. nucleatum (ATCC25586) and A. actinomycetemcomitans (ATCC33384) grown in MHA medium. Each medium was incubated in an anaerobic incubator and its inhibitory power was measured using calipers.
Results: Based on the results of SEM analysis at 1000X magnification, tetracycline can be seen on the surface of chitosan, while FTIR analysis shows compatibility between chitosan and tetracycline. The results of the ANOVA test showed significant differences in the mean diameter of the inhibition zone across groups against periodontal pathogenic bacteria. The average diameter of the chitosan chip 0.7% tetracycline inhibition zone against P. gingivalis is 14.75 ± 0.75 (strong), while against A. actinomycetemcomitans bacteria 25.67 ± 2.082 (very strong) and
F. nucleatum 35.33 ±0.764 (very strong).
Conclusion: chitosan chips 0.7% tetracycline are effective in inhibiting the growth of A. actinomycetemcomitans, P. gingivalis and F. nucleatum bacteria. Chips tetracycline 0.7% is a promising local drug delivery for local therapy of periodontal disease.
