Hepatoprotective Effect Test of Virgin Coconut Oil on Isoniazid and Ripampicin-Induced Hepatotoxicity in Wistar Rats

Impaired liver function is closely related to liver damage. The liver is prone
to damage due to exposure to toxic materials. Rifampicin and isoniazid are first-line
anti-tuberculosis drugs but are also the most potent hepatotoxic compounds. Virgin
Coconut Oil (VCO) has antioxidant compounds that function to counteract free
radicals. The antioxidants contained in Virgin Coconut Oil can function as
hepatoprotective. This study aims to analyze the hepatoprotective effect of virgin
coconut oil in rats induced by isoniazid and rifampicin.
This study used 25 Rattus Norvegicus rats which were divided into 5 groups:
normal controls were only given aquadest, positive controls were given Curcuma
Force 3.6 mg/KgBW/day after 1 hour followed by isoniazid 100 mg/KgBW/day
and rifampicin 100 mg /KgBW/day, negative control was given isoniazid 100
mg/KgBW/day and rifampicin 100 mg/KgBW/day, treatment group 1 was given
VCO 1 ml/KgBW/day after 1 hour followed by isoniazid 100 mg/KgBW/day and
rifampicin 100 mg/KgBW/day, and treatment 2 was given VCO 3 ml/KgBW/day
after 1 hour, followed by isoniazid 100 mg/KgBW/day and rifampicin 100
mg/KgBW/day. The treatment was carried out every day for 14 days, then surgery
and blood sampling were performed. Blood sample for examination of liver damage
biomarkers (TB, ALT, AST) as well as the liver for macroscopic and microscopic
examination. Data were analyzed using one way-ANOVA and then using Post Hoc
Turkey HSD.
The results showed that the changes in ALT, AST and total bilirubin values
in the test animals showed statistically significant differences (p<0.05) compared to
negative controls and normal controls as well as histopathological images of the
liver in the form of fatty degeneration, hydropic degeneration, necrosis and
congestion. Based on the results of the study it was concluded that VCO could act
as a hepatoprotective. VCO at a dose of 1 ml/KgBB/day and VCO at a dose of 3
ml/KgBW/day can reduce ALT, AST and total bilirubin values and can reduce liver
damage.
