dc.description.abstractFixed orthodontic appliances have components with designs that are difficult to clean, leading to plaque accumulation. One of the methods to inhibit bacterial growth is the use of mouthwash. The ant nest plant (Myrmecodia pendans) demonstrates effectiveness in inhibiting Staphylococcus aureus bacteria. The aim of this research is to compare the number of Staphylococcus aureus colonies after immersing brackets in ant nest tuber extract mouthwash, Chlorhexidine gluconate 0.2%, and artificial saliva. The design of this research is a post-test only with a control group design. The study's sample consists of 25 Ormco stainless steel edgewise slot 0.22-inch brackets central incisor. Samples are placed in tubes containing bacterial suspension and incubated for 24 hours. Then, five brackets are immersed for one hour in tubes containing ant nest tuber extract mouthwash at concentrations of 0.8%, 1.6%, 3.2%, Chlorhexidine gluconate 0.2%, and artificial saliva. The samples are removed and immersed in 2 ml Nutrient Broth, then cultured on Plate Count Agar media, incubated for 24 hours, and colonies are counted. The research results indicate the mean number of Staphylococcus aureus bacterial colonies with the 0.8% concentration of ant nest tuber extract mouthwash is 702.8±92.8, the 1.6% concentration is 392.4±31.6, the 3.2% concentration is 129±41.8, Chlorhexidine gluconate 0.2% is 23±6, and artificial saliva is 1074.2±86. Based on the conducted research, it can be concluded that there is significant difference in the number of bacterial colonies after immersion with ant nest tuber extract mouthwash, Chlorhexidine gluconate 0.2%, and artificial saliva.en_US
