Dysplasia is defined as a pre-cancerous lesion of stratified squamous epithelium characterized by cellular atypia and loss of maturation and stratification. Gotu Kola (Centella asiatica L.) is a natural substance that can be used as a chemopreventive agent. This research aims to determine the effectiveness of methanol extract of Gotu Kola leaves (Centella asiatica L.) at doses of 50 mg/kg BW and 100 mg/kg BW on TNF-α expression in DMBA-induced dysplasia. This laboratory experimental study involved 30 Wistar rats (Rattus norvegicus) divided into three groups. In Group 1, the sample was given methanol extract of Gotu Kola leaves at a dose of 50 mg/kg BW and induced with DMBA three times a week. Group 2 received only DMBA three times a week, while Group 3 was given methanol extract of Gotu Kola leaves at a dose of 100 mg/kg BW and induced with DMBA three times a week. The evaluation of TNF-α expression in this study used the IRS score. The results of TNF-α expression measurements were analyzed using the Kruskal-Wallis test and followed by the Mann-Whitney test. The study found no significant results in the administration of methanol extract of Gotu Kola leaves. There was no effectiveness in the administration of methanol extract of Gotu Kola (Centella asiatica L.) leaves at doses of 50 mg/kg BW and 100 mg/kg BW on TNF-α expression in DMBA-induced dysplasia.
