dc.description.abstractOtotoxic drugs such as cisplatin can cause permanent damage to the auditory system, one of which is by mediating pro-inflammatory cytokines through activation of nuclear factor kappa B (NF-κB). A number of studies have also reported that curcumin has many benefits, such as antioxidant and anti-inflammatory through the regulation of certain transcription factors such as NF-κB. The use of OAE allows for earlier indications of cochlear damage. The Enzyme-Linked Immunosorbent Assay (ELISA) test represents a variety of analytical methods because it has several advantages over other analytical methods. It has also been widely used to detect antibodies to viruses and autoantigens in serum or whole blood. This research to prove curcumin as a herbal therapy to prevent ototoxicity caused by cisplatin, assessed by examining OAE and determining the effect of curcumin administration on NF-kB levels in the blood of Rattus norvegicus. This study used 24 mice divided into 4 groups. The first group was not given the treatment. The second, third and fourth groups were given a single dose cisplatin at a dose of 8 mg/kgBW (day 0), with the second group without curcumin, the third group given curcumin at a dose of 150 mg/KgBW and the fourth group at a dose of 300 mg/KgBW. Curcumin was given from day 0 to day 7. OAE examinations were carried out on day 0 and day 7. NF-κB levels were examined using the ELISA method on the 7th day after the mice were terminated. Cisplatin is capable of causing cochlear damage, which is indicated by a decrease in SNR values. On the other hand, curcumin at optimal doses was able to improve it, as indicated by an increase in SNR values, although there was no significant difference in NF-κB levels in the blood.en_US
