dc.description.abstractBackground: Benign Prostate Hyperplasia (BPH) is a disease that often occurs in elderly men and has a serious impact on public health. Obesity is one of the main factors that increase the risk of chronic non-communicable diseases such as heart problems, type 2 diabetes and BPH. Objective: To analyze the effect of extracts of puguntano extract (Picria fel-terrae Merr.) on PI3K, marker of inflammation, oxidative stress, PSA, prostate index and prostate tissue in obese Wistar rats with BPH. Methods: This study used an experimental method which was divided into 5 groups, namely the normal group/K1, negative control group/K2 (obese + BPH), positive control group/K3 (obese + BPH + finasteride 1 mg/KgBW), treatment group 1/K4 (obese + BPH + puguntano extract 100 mg/KgBW) and treatment group 2/K5 (obese + BPH + puguntano extract 200 mg/KgBW). A high-fat diet was given for 12 weeks and testosterone 10 mg/KgBW was injected subcutaneously for 4 weeks. Puguntano extract were administered orally for 10 days. Then, the examination of PI3K, interleukin-6, VEGF and PSA using the ELISA method and calculation of the prostate index value and prostate histopathological examination. Data analysis was carried out using the one-way ANOVA test with normal distribution and Kruskal-Wallis test if the distribution was not normal. Results: The results showed a significant interleukin-6 levels (p=0.024), PSA levels (p<0.001), prostate index values (p<0.001) and significant improvement of prostate histopathological examination in stromal and epithelial cells (p<0.001). Administration of puguntano extract (Picria fel-terrae Merr.) didnot affect PI3K levels (p=0.069) and VEGF levels (p=0.274) Conclusion: This study showed that puguntano leaf extract could reduce marker of inflammation, improve PSA levels, prostate index and prostate histopathology of obese Wistar rats with BPH.en_US
