Determination of Total Phenols, Total Flavonoids and Antioxidant Activity by ABTS Method of Ethanol Extract of Leaves of Sandoricum koetjape (Burm.f.) Merr
Abstract
Background: Sandoricum koetjape leaves from the Melliaceae family contain
active substances such as flavonoids, tannins, phenols and other secondary
metabolites that can play an active role as antibacterial, antimicrobial, antioxidant,
anti-inflammatory, anticholesterol, antifungal and anticancer.
Objective: To determine the levels of phenols, flavonoids, and antioxidant
activity using the ABTS method from the ethanol extract of Sandoricum koetjape
(Burm.f.) Merr leaves.
Method: Powdered simplisia was macerated with 96% ethanol solvent and
concentrated using a rotary evaporator. Then the ethanol extract of Sandoricum
koetjape leaves was tested for total phenol content by colorimetric method in UVVis
spectrophotometry using Folin-Ciocalteu reagent with gallic acid standard,
total flavonoid content was carried out by adding AlCl3 reagent with quercetin
standard, and antioxidant activity test against ABTS at 731 nm wavelength using
UV-Vis spectrophotometer.
Results: Phytochemical screening of Sandoricum koetjape leaves showed the
presence of flavonoids, glycosides, saponins, tannins and steroids/triterpenoids.
The results showed that the levels of total phenols and total flavonoids contained
in the ethanol extract of Sandoricum koetjape leaves were 21,8067 mg GAE/g
sample and 10,4821 mg QE/g sample, respectively, and the antioxidant activity
test of the ethanol extract of Sandoricum koetjape leaves using the ABTS (2,2'-
azino-bis(3- ethylbenz-thiazoline-6-sulfonic acid) method showed an IC50 value
of 66,2144±0,0104 μg/ml with strong antioxidant activity category.
Conclusion: The results concluded that the ethanol extract of Sandoricum
koetjape leaves has high phenol, flavonoid, and antioxidant content.
