BACKGROUND: Turmeric has many health benefits. The largest components in turmeric are curcumin, demethoxycurcumin, and bisdemethoxycurcumin which have antioxidant, hepatoprotective, cardioprotective, antifungal and anticancer activities. The UHPLC method was used to examine the levels of curcumin, demethoxycurcumin, and bisdemethoxycurcumin in turmeric ethanol extract.
OBJECTIVE: To examine the levels of curcumin, demethoxycurcumin, and bisdemethoxycurcumin in turmeric ethanol extract using UHPLC method and to determine whether UHPLC method and conditions used met the validation requirements.
METHODS: Analysis using the UHPLC method used a mobile phase of 0.25% acetonitrile-acetic acid (50:50) with a flow rate of 0.8 ml/minute, column temperature of 35℃ with a wavelength of 425 nm, injection volume of 10 μl and a C18 column.
RESULTS: The levels of curcumin were 170.3129 mg, demethoxycurcumin 55.0668 mg, and 33.43265 mg bisdemethoxycurcumin. In calibration, correlation coefficient for curcumin was 0.9999, demethoxycurcumin 0.9998, bisdemethoxycurcumin 0.9998, %recovery of curcumin 97.9572%, demethoxycurcumin 96.2547%, bisdemethoxycurcumin 95.6522%, %RSD of curcumin 1.4823%, demethoxycurcumin 1.8626%, bisdemethoxycurcumin 1.6373%. LOD of curcumin 1.7590 ppm, demethoxycurcumin 0.3976 ppm, bisdemethoxycurcumin 0.1061 ppm and LOQ curcumin 4.1408 ppm, demethoxycurcumin 1.2050 ppm, bisdemethoxycurcumin 0.3214 ppm.
CONCLUSION: The levels of the compounds curcumin, demethoxycurcumin, and bisdemethoxycurcumin in the ethanol extract of turmeric were curcumin 170.3129 mg, demethoxycurcumin 55.0668 mg, and 33.43265 mg bisdemethoxycurcumin. The method used meets the validation test requirements.
