Isolasi Fragmen Gen pup1 sebagai Gen Ketahanan terhadap Penyakit Blas pada Padi Sigudang (Oryza Sativa L.) dari Kabupaten Mandailing Natal
Isolation of pup1 Gene Fragment as a Resistance Gene to Blas Disease in Sigudang Rice (Oryza Sativa L.) from Mandailing Natal Regency
Abstract
One of the common diseases that attack rice plants in Indonesia is blast disease. It is caused by the fungus Pyricularia oryzae. The characteristic of this disease is characterized by the presence of necrotic, so because of that. it disrupts the absorption of nutrients and the growth of rice plants. Mandailing Natal Regency has local rice varieties that are resistant to blast disease. One of them is Sigudang rice because it has genes that are resistant to blast disease, one of which is the pup1 gene. The pup1 gene is found on chromosome 12 and plays a role in the capture of phosphorus (P) in rice plants. The pup1 gene encodes a dirigent-like protein for the biosynthesis process of lignin which affects the hardness of plant cell walls so as to prevent infection of plants. This study aims to isolate the gene fragment and sequencing of pup1 gene in Sigudang rice. The isolated pup1 gene fragment was amplified with specific primers and sequencing analysis was conducted. Genomic DNA from Sigudang rice has been successfully isolated and pup1 specific primers successfully amplified pup1 gene fragment measuring 650 bp. After sequencing and analyzed with BLASTn, it was found that the candidate fragment of the pup1 gene from Sigudang rice had 98.92% similarity with the pupk20-2 gene in Oryza sativa cultivar Murungakayan and Oryza sativa cultivar Suduheenati. Dendrogram analysis showed that the nucleotide sequence of the pup1 gene fragment of Sigudang rice clustered with the nucleotide sequence of the pup1 gene in other rice with a genetic distance of 0.262.
