The research of the chlorophyll and stomata levels on Ashoka Tree (Polyalthia longifolia) from several locations at Sumatera Utara exposed to lead (Pb), was conducted since January until June 2023 at Laboratorium Fisiologi Tumbuhan dan Kultur Jaringan, Laboratorium Mikrobiologi, Fakultas Matematika dan Ilmu Pengetahuan Alam and UPT. Laboratorium Penelitian Terpadu, Universitas Sumatera Utara. The purpose of this research was to compare chlorophyll levels, the number and length of stomata, the stomata index, the thickness and the leaf area of the ashoka Tree (Polyalthia longifolia) base on the lead absoption levels from several locations at Sumatera Utara, as well as to analyzing the correlation of the stomata index with leaf area. The design used in this study was Randomized Block Design (RAK) and data analysis used One-Way ANOVA then continued by Duncan's test (DMRT). The results of lead content analysis were done by using the wet ashing method using an Atomic Absorption Spectrophotometer (AAS). The result show the average lead levels from the highest to the lowest found at Jalan Pulau Nusa Barung (0,75 mg/L), Jalan Gatot Subroto (0,67 mg/L), Jalan Letjen Jamin Ginting (0,33 mg/L), and Jalan Dr. Mansyur (0,26 mg/L). The chlorophyll content analysis by extraction using 80% acetone and gain the average total of chlorophyll levels from the highest to the lowest from Jalan Letjen Jamin Ginting (2,53 mg/L), Jalan Dr. Mansyur (2,42 mg/L), Jalan Gatot Subroto (2,17 mg/L), and Jalan Pulau Nusa Barung (1,95 mg/L). The average number of ashoka leaf stomata from the highest to the lowest is at Jalan Pulau Nusa Barung (12,51 stomata/mm2), Jalan Gatot Subroto (12,13 stomata/mm2), Jalan Letjen Jamin Ginting (11,67 stomata/mm2), and Jalan Dr. Mansyur (6,10 stomata/mm2). The average size of stomata glodokan pole leaves from the highest to the lowest is from Jalan Pulau Nusa Barung (53,52 μm), Jalan Gatot Subroto (52,21 μm), Jalan Dr. mansyur (45,24 μm), and Jalan Letjen Jamin Ginting (44,59 μm). The average stomata index of glodokan pole leaves from the highest to the lowest is from Jalan Pulau Nusa Barung (65,66 %), Jalan Letjen Jamin Ginting (62 %), Jalan Gatot Subroto (61,33 %), and Jalan Dr. Mansyur (54 %). The average thickness of glodokan pole leaves from the highest to the lowest find at Jalan Pulau Nusa Barung (0,48 mm), Jalan Gatot Subroto (0,46 mm), Jalan Letjen Jamin Ginting (0,24 mm), and Jalan Pulau Dr. Mansyur (0,18 mm). The last is the average area of glodokan pole leaves from the highest to the lowest find at Jalan Pulau Nusa Barung (55,6 cm2), Jalan Gatot Subroto (50 cm2), Jalan Dr. Mansyur (42,3 cm2), and Jalan Letjen Jamin Ginting (38,3 cm2).en_US
