Isolation and Identification of Flavonoid Compounds from Tamarind Leaves (Tamarindus Indica L.)
Abstract
Isolation and identification of flavonoid compounds from Tamarind leaves
(Tamarindus indica L) have been carried out. Isolation was carried out using a
maceration extraction stage with methanol solvent. The methanol extract was
concentrated and then dissolved in distilled water. The distilled water solution is
extracted and partitioned with ethyl acetate, the ethyl acetate layer is separated and
concentrated. The concentrated extract of ethyl acetate was dissolved with methanol
and partition extracted with n-hexane. The methanol layer is evaporated until all the
methanol has evaporated. The concentrated methanol extract was analyzed by TLC,
then separated by column chromatography with a silica gel stationary phase and
chloroform: ethyl acetate solvent in the ratio (90:10; 80: 20; 70: 30; 60: 40; 50: 50;
40: 60; 30 :70;20:80;10:90) v/v as the mobile phase. The fourth fraction was chosen
to be purified by thin layer chromatography with the stationary phase kieselgel 60 F254
and chloroform: ethyl acetate (60:40) v/v as the mobile phase. The pure compound
obtained was in the form of a yellow amorphous solid of 5.5 mg with a value of Rf=0.52
using chloroform eluent: ethyl acetate (60:40) v/v. based on the results of UV-Vis
Spectrum analysis, it shows band I (λ max) 370 nm and band II 280 nm. The FT-IR
spectrum shows the functional groups OH, C-H sp3, Stretching, C=O ketone, C=C
aromatic, C-H sp3 bending, C-O, C-O-C, C-H sp2 bend out-of-plane with wave
numbers respectively 3429.43 cm-1, 2956.87 cm-1, 1723.29 cm-1, 1598.99 cm-1,1381.03
cm-1, 1286.52 cm-1, 1122.57 cm-1, 742.59 cm-1. The Proton Nuclear Magnetic
Resonance (1H-NMR) spectrum shows H-6, H-8, H-2', H-6' and OCH3 protons.
Spectroscopic data shows that the isolated compound is a flavonoid compound in the
flavonol group.
