Induction of Mangosteen Callus (Garcinia mangostana L.) with the Addition of BAP (Benzyl Amino Purine) and NAA (Naphthalene Acetic Acid) In Vitro
Abstract
The aim of the study was to determine the effect of the concentration of the combination of ZPT BAP and NAA and the interaction of both on the growth of mangosteen explants (Garcinia mangostana L.). This research was carried out at the Plant Biotechnology Laboratory, Faculty of Agriculture, University of North Sumatra, from January to April 2024. This study used the Complete Randomized Design (RAL) method with 2 treatment factors. The first factor is Benzyl Amino Purine (BAP) which consists of 0 mg/L, 3 mg/L, and 6 mg/L. The second factor is Naphthalene Acetic Acid (NAA) which consists of 0 mg/L, 3 mg/L, and 6 mg/L. The parameters observed are the percentage of living explant, the day the callus appears, the texture of the callus, and the color of the callus. The results showed that the addition of ZPT, BAP and NAA had no real effect on the percentage of living explants. The addition of ZPT, BAP and NAA had a real effect on the day the callus appears. The addition of ZPT, BAP and NAA has a good effect on the texture and color of the callus. Based on the results of the study, it was shown that the best treatment was at a concentration of 3 mg/L BAP and NAA on the growth of mangosteen explant in vitro.
