Antioxidant Activity Test of Ethanol Extract, N-Hexane Fraction and Ethyl Acetate Fraction of Rantiti Leaf (Cipadessa baccifera (Roxb. Ex Roth.) Miq.) DPPH
Abstract
Background: Rantiti (Cipadessa baccifera (Roxb. Ex Roth) Miq.) belongs to the Meliaceae family having secondary metabolite compounds in the form of Flavonoids, Phenolics, Steroids, Triterpenoids, Glycosides, Alkaloids and Saponins. The compounds in Rantiti leaves are compounds that have the potential to be medicinal ingredients. Flavonoids are one of the powerful antioxidant compounds and are commonly found in plants. Fractionation is carried out to separate compounds based on their polarity so that purer compounds are obtained.
Objective: To determine the antioxidant activity of ethanol extract, n-hexane fraction and ethyl acetate fraction of trationite leaves by DPPH method.
Methods: Simplicia powder was subjected to a characteristic test and phytochemical screening and then extracted by maceration with 96% ethanol solvent and then the extract was fractionated in a liquid-liquid manner with n-hexane and ethyl acetate solvents. Ethanol extracts and fractions obtained in the antioxidant activity test by UV-Vis spectrophotometry by DPPH method at a wavelength of 515,5 nm with an Operating Time of 41 minutes
Results: The examination of the characteristics of simplicia of rantiti leaves obtained a moisture content of 7,27%, a water-soluble juice content of 50,39%, an ethanol-soluble juice content of 13,69%, a total ash content of 6,16% and an acid insoluble ash content of 1,34%. The results of phytochemical screening of simplicia leaf of the trelliti contain alkaloids, flavonoids, tannins, saponins, glycosides, steroids/triterpenoids. The results of the antioxidant activity test of ethanol extract of rantiti leaves showed an IC₅₀ value of 54,0935±0,1597 μg/ml in the strong category, The IC₅₀ value of the n-hexane fraction of 529,8362±24,1563 μg/ml is in the weak category, the IC₅₀ value of the ethyl acetate fraction is 45,0267±0,2748 μg/ml is in the very strong category and the quercetin has an IC₅₀ value of 2,4315±0,0137 μg/ml in the very strong category.
Conclusion: Ethanol extract of Rantiti leaf has strong antioxidant activity, the n-hexane fraction of Rantiti leaf has weak antioxidant activity and the ethyl acetate fraction of Rantiti leaf has very strong antioxidant activity.
